Description

Retatrutide is a synthetic 39-amino-acid peptide. It was designed in a laboratory and is not a molecule the body produces — the sequence borrows from natural gut hormones but has been deliberately modified. What makes it structurally unusual is that it was engineered to engage three different receptors, where earlier peptides in the same family engage one or two.

Cells in your gut, pancreas, liver and brain carry receptors that respond to hormones released around the time you eat. Retatrutide binds three of them. Two — the GLP-1 receptor and the GIP receptor — belong to what physiologists call the incretin system, which helps coordinate insulin release and how quickly the stomach empties. The third, the glucagon receptor, sits mostly on liver cells and is involved in mobilising stored energy. Hitting all three at once is the entire design intent, and characterising what that combination does is what the ongoing Phase 3 programme exists to measure.
